Derived from Bacillus subtilis, Nattokinase (CAS 133876-92-3, EC 3.4.21.62) is a food-grade alkaline serine protease refined and extracted through fermentation. It is an effective fibrinolytic enzyme that can break down blood clots by directly hydrolyzing fibrin and plasmin substrates. As a promising material, it is also widely used in the research of cardiovascular diseases.

Definition of Enzyme Activity

Nattokinase reacts with fibrin, hydrolyzing the peptide bonds of fibrin. The hydrolysis reaction causes a change in the absorbance of the solution at 275nm under ultraviolet light. One activity unit (1FU) refers to the amount of enzyme consumed when the absorbance value increases by 0.01 at 275nm.

Functioning Principle and Dosage

The main component of thrombus is fibrin. It is a stable and sticky protein composed of polypeptide chains. Fibrinolysin can cut the peptide bonds of fibrin, breaking it down into small molecules, thus dissolving the thrombus. The main mechanism of action of nattokonase is to activate plasminogen, converting it into fibrinolysin, thereby decomposing the fibrin in the thrombus and dissolving the thrombus.


The thrombolytic effect of nattokonase has strong selectivity and specificity. It can only act on the fibrin in the thrombus and will not affect the healthy blood vessel wall. Therefore, nattokinase is considered a safe and effective thrombolytic agent and has been widely used in the clinical treatment of cardiovascular diseases.


For the prevention of cardiovascular diseases, the recommended daily intake is 2000FU; for patients with cardiovascular and cerebrovascular diseases, the recommended daily intake is 4000-6000FU.
